Advantages of MES buffer

Preparation method of MES buffer:

1. Prepare 800 mL of dH2O in a suitable container.

2. Add 97.62 grams of MES free acid to the dH2O solution.

3. The initial pH value of 0.5M MES solution is 3.23. For 1 L of 0.5M MES, 13.6 ml of 10N NaOH is required to adjust the pH to 6.0;

4. Add dH2O until the volume is 1L.

Storage: MES solution can be stored for several months at 2-8 ºC. It is recommended to use a 0.2 micron filter membrane for filtration and sterilization. (Substances containing sulfate ions are not easily sterilized at high temperatures) After high-temperature sterilization, MES will show a light yellow color. It is not clear what the light yellow degradation product is, but the pH will not change significantly.
